One of Sebia's most unique kits, the IF Penta assay is designed as a screening immunofixation. Penta is considered a true and complete immunofixation assay. It contains all antisera utilized in a traditional immunofixation; however, it streamlines the test procedure thus expediting the processing of large workloads. All negative results can be immediately reported with any positive results being further identified with the traditional IF assay. For laboratories with large immunofixation workloads with a high percentage of negative or normal results, the Penta assay can provide a tremendous cost and labor savings.
As with the IF assay, the sample is first electrophoresed; however, with Penta neat sample is used saving a dilution step. The Dynamic Mask is also employed in this assay to greatly simplify antisera application and reducing manual pipetting. The HydraPLUS instrument can also be used to completely eliminate antisera pipetting. In this case, a carefully formulated fixative and pentavalent mixture of the traditional 5 antisera (IgG, IgA, IgM, Kappa and Lambda) are applied. Only two tracks are required verses the traditional six so more samples can be processed simultaneously speeding workflow. Again, no time-consuming press drying steps are required with this assay! A simple blotting step removes all protein that has not combined with the applied antisera allowing any abnormal proteins to be easily identified. With assay sensitivity, band resolution and gel clarity equivalent to IF, the IF Penta assay correlates well to Sebia's immunofixation assay.
